GH001 Shows Rapid Relief for Postpartum Depression in Early Trial

GH001 and Postpartum Depression: Promising Early Data

The field of perinatal psychiatric treatment has expanded rapidly. New options like brexanolone and zuranolone have diversified care for postpartum depression. Researchers are now exploring psychedelics as potential solutions.

A phase 2a trial published in The Journal of Clinical Psychiatry evaluated GH001. This is a synthetic inhalable formulation of methylenedioxymethamphetamine (5-MeO-DMT). The study targeted adults with moderate or severe postpartum depression. Results indicate rapid antidepressant effects. Significant symptom reduction was observed within hours of administration. However, these data are preliminary and require confirmation in larger controlled studies.

What exactly is GH001? It is an investigational compound found naturally in certain plants and the venom of the Colorado river frog. The synthetic formulation is designed for clinical inhalation. The mechanism involves serotonin receptor activation. It has higher affinity for the 5-HT1A subtype than 5-HT2A. Its short plasma half-life means acute psychoactive effects last only 20-25 minutes per dose. These effects include intense changes in consciousness and perception. Due to the intensity, administration requires rigorous monitoring.

Study Design

Prior clinical trials of GH001 in treatment-resistant depression reported rapid antidepressant effects and an acceptable safety profile. This phase 2a study is the first investigation of GH001 for postpartum depression.

This open-label, proof-of-concept phase 2a trial was conducted in the UK and the Netherlands. It included 10 women aged 18 to 45. All had a diagnosis of major depressive disorder with peripartum onset. This was confirmed via the Mini-International Neuropsychiatric Interview. At dosing, participants were more than four weeks postpartum. Screening occurred within 12 months postpartum. All presented moderate to severe depressive symptoms. Baseline scores were at least 28 on the Montgomery-Asberg Depression Rating Scale (MADRS).

Results

GH001 was administered under medical supervision using an individualized dosing regimen: - Participants initially received a 6 mg inhaled dose. - If no intense psychoactive effect was experienced and the prior dose was safe and well-tolerated, additional 12 mg and 18 mg doses could be given. - A maximum of three doses were administered in a single day, with approximately one hour between doses.

The study did not include planned psychotherapy before, during, or after dosing. However, administration occurred under clinical supervision. Changes in depressive symptom severity were measured by total MADRS score from baseline to day 8. Antidepressant response was defined as at least a 50% reduction in MADRS score. Remission was defined as a MADRS score of 10 or lower.

Ten participants were enrolled. The mean baseline MADRS score was 36.7 (SD = 4.8), indicating considerable depressive symptom severity.
